Potent weapons for the oncology arsenal
نویسنده
چکیده
Antibody Drug Conjugates (ADCs) are monoclonal antibodies attached to biologically active drugs by chemical linkers with selectively labile bonds. By combining the unique targeting of monoclonal antibodies with the cancer killing ability of cytotoxic drugs, ADCs will permit sensitive discrimination between healthy and diseased tissue. Advances in coupling antibodies to cytotoxic drugs will permit greater control of drug pharmacokinetics and significantly improve delivery to target tissue. Potent new anticancer drugs can now be used to target cancers while minimizing exposure of healthy tissue. Promising clinical results will continue to drive clinical development of this therapeutic class. Manufacturing highly potent biopharmaceuticals presents a series of unique engineering and chemistry challenges that have resulted in Lonza designing and building a new dedicated manufacturing plant in Visp, Switzerland. By leveraging experience in biopharmaceuticals, small molecule drug process development and scale-up, this class of high potency biologicals can now be manufactured to support both clinical development and licensure.

Past, Present, and Future of Molecular and Cellular Oncology
In the last 20 years, the field of cellular and molecular oncology has been born and has moved its first steps, with an increasingly rapid pace. Hundreds of oncogenic and oncosuppressive signaling cascades have been characterized, facilitating the development of an ever more refined and variegated arsenal of diagnostic and therapeutic weapons. Furthermore, several cancer-specific features and p...
متن کاملProjected Costs of U.S. Nuclear Forces, 2017 to 2026
Nuclear weapons have been a cornerstone of U.S. national security since they were developed during World War II. In the Cold War, nuclear forces were central to U.S. defense policy, resulting in the buildup of a large arsenal. Since that time, nuclear forces have figured less prominently than conventional forces, and the United States has not built any new nuclear weapons or delivery systems fo...
متن کاملNuclear winter revisited with a modern climate model and current nuclear arsenals: Still catastrophic consequences
[1] Twenty years ago, the results of climate model simulations of the response to smoke and dust from a massive nuclear exchange between the superpowers could be summarized as ‘‘nuclear winter,’’ with rapid temperature, precipitation, and insolation drops at the surface that would threaten global agriculture for at least a year. The global nuclear arsenal has fallen by a factor of three since t...
متن کاملVerifying Nuclear Disarmament
Commentators differ on whether nuclear disarmament would be desirable, but many argue that disarmament is impractical because it could not be verified. Three reasons are often offered for such pessimism. First, nuclear weapons are small and difficult to detect, and one could not be sure that a few weapons had not been hidden away. Second, nuclear weapons are so destructive that a mere handful w...
متن کاملMarketed nonsteroidal anti-inflammatory agents, antihypertensives, and human immunodeficiency virus protease inhibitors: as-yet-unused weapons of the oncologists’ arsenal
Experimental data indicate that several pharmacological agents that have long been used for the management of various diseases unrelated to cancer exhibit profound in vitro and in vivo anticancer activity. This is of major clinical importance, since it would possibly aid in reassessing the therapeutic use of currently used agents for which clinicians already have experience. Further, this would...
